AVALIAÇÃO DE TOXICIDADE AGUDA: ESTRATÉGIAS APÓS A “ERA DO TESTE DL50“

Abstract

RESUMO: O presente trabalho discute a avaliação da toxicidade aguda sistêmica após a eliminação do teste da DL 50 (dose letal mediana). Os três testes alternativos em animais empregados hoje, Dose Fixa, Toxicidade Aguda de Classe e o Teste " Up and Down " , trouxeram significante melhora para o bem-estar animal. Embora a eliminação total do uso de animais ainda não seja possível, a citotoxicidade basal in vitro tem sido empregada como adjuvante dos testes em animal para a seleção das doses iniciais reduzindo ainda mais o número de animais empregados na avaliação do toxicidade aguda sistêmica. PALAVRAS-CHAVE: toxicidade oral aguda; métodos alternativos de avaliação de toxicidade; citotoxicidade ABSTRACT: The present work discuss about the evaluation of the acute systemic toxicity post deletion of the LD 50 test (median lethal dose). The three alternative animal tests now employed, the Fixed Dose Procedure, the Acute Toxic Class Method and the Up and Down Procedure, give rise to significant improvements in animal welfare. Although, the total replacement of animal tests it is not possible yet, basal cytotoxicity in vitro has been used as adjunct to these alternative animal tests to select initial doses, thus reducing the numbers of animals used during the evaluation of acute systemic toxicity.
