Abstract

A Simple, rapid and precise high performance liquid chromatography analysis for tea catechins was developed. The separation system consisted of a VDSpher Pur 100 C18-E column, a gradient elution system of acetonitrile-methanol-phosphoric acid 0.05% with the ratio 1:49:50 and sodium dihydrogen orthophosphate 20mM at pH 2.5, 1,2ml menit -1 flowrate and an diode array detector. The validation of this method was confirmed from specificity, linearity, precision, limit of detection and limit of quantification. The result exhibited good linearity and accuracy in the range of 0.4 to 47.34mg/kg with coefficient of determination 0.9906. A pretty detection limit of the method for catechin, epigallocathecin gallat, epicatechin are 0.36, 0.51, 0.35mg/kg obtained.. Moreover, the application of the validated HPLC method for Cathecin measurement Gamboeng Tea Superior Clone indicated that the method is suitable and reliable for routine work. Besides, the use of a traceable standard in all experiment assured that the measurement results are traceable internationally. Furthermore, this method can be applied to all kinds of tea and tea products, and it is especially useful for the determination of catechin, which was regarded as being in too low a concentration to detect.
