Abstract

要 旨 生体試料中において透過性の高い近赤外光を用いることは深部イメージングにおいて有用である.最近では,近赤外 光を用いた非線形光学顕微鏡が生体組織の深部イメージングに広く使用されている.しかし,散乱や吸収による光損失 が大きい試料の深部観察は困難である.その原因は,焦点面以外で発生する背景光にある.本解説では,非線形光学顕 微鏡における背景光の発生機構と背景光抑制技術を紹介する. キーワード:非線形光学顕微鏡,深部イメージング,超解像イメージング,蛍光イメージング Abstract Near-infrared light that provides maximum optical transparency in biological systems is a powerful tool to image deeper inside biological samples. Recently, the use of nonlinear optical microscope using near-infrared light has been widely spread in deep tissue imaging. Nevertheless, it is difficult to observe a highly scattering sample at a deeper penetration depth. This is because background noise is generated from out-of-focus regions. In this review, we explain the mechanism of background generation in nonlinear optical microscopy and introduce some methods to suppress the background noise.
