RFLP and VNTR Markers Isolated from the Genomic DNA of Rainbow Trout and Cytogenetic Mapping by Fluorescence in Situ Hybridization

Abstract

To obtain restriction fragment length polymorphism (RFLP) and variable number of tandem repeat (VNTR) markers from the rainbow trout genome, we constructed a genomic cosmid library from the RTG-2 cell line. Cosmid DNAs were prepared from 25 clones and examined to detect DNA polymorphisms. Three conventional RFLPs and 5 VNTR markers were identified. One of the VNTR markers (RVF9301) consisted of repeat units of a minisatellite tandem repeat of 52 bp in length. In a primary study for cytogenetic mapping, we succeeded in localizing these markers on rainbow trout chromosomes by fluorescence in situ hybridization. Our results suggest these 8 clones will serve as useful genetic markers for linkage analysis of genetic traits in fisheries science.
