Abstract
All single-stranded RNA (ssRNA) virus-infected plants produce double-stranded RNA (dsRNA) during their replication cycle. dsRNAs are more stable than its counterpart ssRNA. Many protocols are available for the isolation ofdsRNA from virus-infected plants that are used as template for cDNA synthesis followed by PCR assay. In this chapter, suitable protocol for the isolation of ds RNA from plant sources is discussed.
