(2021) Aktivitas antioksidan dan komponen bioaktif hidrolisat protein jeroan ikan kakap putih (Lates calcalifer) Warta IHP, 38(1),70-78 Halaman | 70 ABSTRAK: Jeroan ikan kakap putih (Lates calcalifer) merupakan limbah padat dari industri pengolahan ikan yang belum dimanfaatkan secara optimal sehingga sangat berpotensi untuk dijadikan produk hidrolisat protein. Penelitian ini bertujuan untuk menentukan senyawa antioksidan dan komponen bioaktif dari hidrolisat protein jeroan ikan kakap putih. Metode yang digunakan adalah optimasi konsentrasi enzim, waktu hidrolisis, dan pH; analisis proksimat, asam amino, aktivitas antioksidan, dan uji komponen bioaktif. Kondisi aktivitas antioksidan tertinggi hidrolisat protein jeroan kakap putih yaitu pada konsentrasi enzim 0,1%, waktu hidrolisis 4 jam, dan pH 7. Produk yang dihasilkan mengandung kadar air (6,83%), abu (5,18%), protein (80,88%), lemak (0,78%), dan karbohidrat (13,15%). Produk ini terdiri dari 15 macam asam amino, yaitu 8 asam amino esensial dan 7 asam amino non esensial. Asam glutamat merupakan asam amino dengan kadar tertinggi (8,71%), sedangkan asam amino dengan kadar terendah adalah histidina (1,38%). Produk ini menunjukkan aktivitas antioksidannya dengan nilai IC50 sebesar 1.048,40 ppm, dan mengandung 5 dari 9 komponen bioaktif, yaitu flavonoid, karbohidrat, gula pereduksi, peptida, dan asam amino bebas. Kata kunci: antioksidan, asam amino, bioaktif, hidrolisat, protein ABSTRACT: Visceral of barramundi (Lates calcalifer) is waste in the company of fisheries which has not been optimized so that making it potentially made to be the product of protein hydrolysate. The purpose of this study was to determined antioxidant activity and bioactive compound in the protein hydrolysate from visceral of barramundi. The methods used were the optimization enzyme, hydrolysis time, and pH; proximate analysis, amino acids, antioxidant activity, and bioactive components. The condition of the highest antioxidant activity of visceral barramundi protein hydrolysate on the concentration of enzyme 0.1%, with hydrolysis times 4 hours length, and pH 7. The result of product contain water content (6.83%), ash (5.18%), protein (80.88%), fat (0.78%), and carbohydrate (13.15%). This product consists of 15 kinds of amino acids, which that 8 are essential amino acids and 7 are non essential amino acids. Glutamic acid is an amino acid with the highest levels (8.71%), while an amino acid with the lowest levels is histidine (1.38%). This product showed the antioxidant activity with IC50 value amount to 1 048.40 ppm, and containing 5 from 9 bioactive components, among them are flavonoid, carbohydrate, reducing agent of sugar, peptide, and free amino acids.
