Abstract
The incorporation of non-canonicalãminoãcids (ncAAs) into proteins isã po w erful technique used in various research fields. Genetic code expansion (GCE) is the most common way toãchieve this:ã specific codon is selected to be decoded byã dedicated tRNA orthogonal to the endogenous ones. In the past 30 years, great progress has been made to obtain novel tRNA synthetases (aaRSs)ãcceptingã variety of ncAAs with distinct ph y sicochemical properties, to de v elop robust in vitroãssa y s orãpproaches f or codon reassignment. T his spark ed the use of the technique, leading to theãccumulation of publications, from which gatheringãll rele vãnt inf ormation canãppear daunting. Here w e present iNClusive ( https://non- canonical-ãas.biologie.uni- freiburg.de/),ã manually curated, e xtensiv e repository using standardized nomenclature that pro vides organiz ed inf ormation on ncAAs successfully incorporated into target proteinsãs verified by mass spectrometry. Since we focused on tRNA synthetase-based tRNA loading, w e pro vide the sequence of the tRNAãndãaR S used f or the incorporation. Deriv ed from more than 687 peer-re vie w ed publications, it currently contains 2432 entriesãbout 466 ncAAs, 569 protein tãrgets, 50 0ãaRSsãnd 144 tRNAs. We foresee iNClusive will encourage more researchers to experiment with ncAA incorporation thus contributing to the further development of this exciting technique.
