Abstract

In order to develop a reliable and rapid transgenic system for functional study of specific buckwheat gene constructs, two different in planta transformation methods were analyzed: vacuum infiltration and infiltration by syringe. The results indicated that the vacuum infiltration method was much more efficient and can therefore be considered the method of choice for buckwheat transformation. .Za funkcionalnu analizu potencijalnih promotorskih fragmenata gena heljde neophodan je efikasan metod transformacije. Imajuci u vidu da je mikro- propagacija i regeneracija heljde dugotrajan proces, optimizovana su dve metode za in planta transformaciju: vakuum infiltracija i infiltracija pomocu sprica. Odredjivane su optimalne vrednosti za: gustinu kulture bakterija Agrobacterium tumefaciens (koje nose vektor za transformaciju) i jacinu vakuuma. Efikasnost transformacije je pracena pomocu kvantitativnog GUS eseja. Rezultati su pokazali da je vakuum infiltracija efikasniji metod transformacije heljde. .
