Abstract

The present investigation was conducted to elucidate the antidiabetic activity and mechanism of lignan Matairesinol (MAT) in Streptozotocin (STZ)-induced diabetes in Wistar Rats. The MAT was administered to diabetic rats (5, 10, and 15 mg/kg), and various biochemical parameters were studied to ascertain its pharmacological effect. Results suggest that MAT, in a dose-dependent manner, improves the level of plasma blood glucose, blood insulin, body weight, and hepatic enzymes, e.g., hexokinase (HK), Glucose 6-phosphatase (G6Pase), and Fructose 1,6-bisphosphatase (FBPase) in Wistar rats. In flow cytometry analysis, MAT significantly prevented apoptosis of hepatocytes compared to STZ rats. In docking analysis, MAT showed excellent interaction with DPP-4 via the formation of hydrophobic interaction of both phenyl rings with Tyr628 and Phe319 of S1 pocket lined with a lipophilic cavity by residues such as Tyr631, Val656, Trp659, Tyr662, Tyr666. It also showed the formation of H-bonds with Glu167, Arg87, Ser171, and His88. The present study showed a significant antidiabetic and hepatoprotective effect of matairesinol.
