Abstract
Ganoderma applanatum belongs to the group of white-rot fungi, due to a well-developed ligninolytic enzyme system. White-rot fungi have attracted great scientific attention in recent years, especially with respect to their enzymatic potential for the bioremediation of persistent pollutants. Contrary to G. lucidum, which medicinal properties, as well as ligninolytic enzyme system have been extensively studied, enzymatic system of G. applanatum has not been studied yet. Thus, the aim of this study was to analyze the dynamics of laccase, Mn-dependent peroxidase, and versatile peroxidase activity during submerged and solid state cultivation on two selected plant raw materials. Enzyme activity was determined spectrophotometrically after 7, 10 and 14 days of cultivation. The peak of laccase activity (220.14 Ul-1) was noted after 14 days of submerged wheat straw fermentation. Maximum level of Mn-dependent peroxidase (110.91 Ul-1) and versatile peroxidase (116.20 Ul-1) activity was obtained in the medium with oak sawdust after 14 days of submerged cultivation.Ganoderma applanatum pripada grupi gljiva izazivaca bele trulezi zahvaljujuci dobro razvijenom ligninolitickom enzimskom sistemu. Ova grupa organizama privlaci veliku naucnu paznju poslednjih godina s obzirom da poseduju veliki enzimski potencijal za razgradnju tesko razgradivih polutanata. Za razliku od vrste G. lucidum, cija su lekovita svojstva i ligninoliticki enzimski sistem detaljno proucavani, enzimski sistem G. applanatum je jos uvek nedovoljno poznat. Cilj ovih istrazivanja je bio da se analizira dinamika produkcije Mn-zavisne peroksidaze (MnP), verzatil peroksidaze (VP) i lakaze (Lac) u medijumu sa psenicnom slamom ili piljevinom hrasta kao izvorima ugljenika, tokom tecne i cvrste kultivacije. Merenja su vrsena nakon 7, 10 i 14 dana kultivacije. Aktivnost enzima je merena spektrofotometrijski, koriscenjem ABTS-a kao supstrata za lakaze i fenol crvenog za Mn-oksidujuce peroksidaze. Maksimalni nivo aktivnosti MnP (110.9 Ul-1) i VP (116.2 Ul-1) je dobijen u medijumu sa piljevinom hrasta, nakon 14 dana tecne kultivacije. Najveca aktivnost Lac (220.1 Ul-1) je izmerena u medijumu sa psenicnom slamom kao izvorom ugljenika, nakon 14 dana tecne kultivacije. PR Projekat Ministarstva nauke Republike Srbije, br. 173032
