Abstract

Abstract. Indonesia has abundant biodiversity, including red algae (Gracilaria sp.) from the Rhodophyta division which has the potential as a source of bioactive compounds such as antioxidants. This study aims to determine the antioxidant activity and total phenol content produced by ethanol extract of red algae (Gracilaria sp.). This study used maceration extraction method with 96% ethanol solvent to obtain active compounds from red algae simplisia. Antioxidant activity was measured using DPPH (1,1-diphenyl-2-picrylhydrazyl) method through calculation of IC₅₀ value and total phenol content was analyzed by Folin-Ciocalteu method. The results obtained from this study are antioxidant activity obtained with IC₅₀ value of 2,521.2 ppm which is classified as very weak and total phenol content of 1.6729 mg GAE/g extract. Antioxidant activity has a relationship with total phenol content, namely the greater the total phenol content, the stronger the antioxidant strength. Abstrak. Indonesia memiliki keanekaragaman hayati yang melimpah, termasuk alga merah (Gracilaria sp.) dari divisi Rhodophyta yang berpotensi sebagai sumber senyawa bioaktif seperti antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dan kadar fenol total yang dihasilkan oleh ekstrak etanol alga merah (Gracilaria sp.). Penelitian ini menggunakan metode ekstraksi maserasi dengan pelarut etanol 96% untuk memperoleh senyawa aktif dari simplisia alga merah. Aktivitas antioksidan diukur menggunakan metode DPPH (1,1-difenil-2- pikrilhidrazil) melalui perhitungan nilai IC₅₀ dan kadar fenol total dianalisis dengan metode Folin-Ciocalteu. Hasil yang didapatkan dari penelitian ini adalah aktivitas antioksidan diperoleh dengan nilai IC₅₀ sebesar 2.521,2 ppm yang tergolong sangat lemah dan kadar fenol total sebesar 1,6729 mg GAE/g ekstrak. Aktivitas antioksidan mempunyai hubungan dengan kadar fenol total, yaitu semakin besar kadar fenol total maka, kekuatan antioksidannya semakin kuat.
