Abstract
We report the synthesis and characterization of a novel nanoparticleformulation designed for skin penetration for the purpose of skinimaging. Solid lipid nanoparticles (SLNs), a drug delivery vehicle, wereused as the matrix for targeted delivery of peroxide-sensitivechemiluminescent compounds to the epidermis. Luminol and oxalate werechosen as the chemiluminescent test systems, and a formulation wasdetermined based upon non-toxic components, lotion-like properties, andlongevity / visibility of a chemiluminescent signal. The luminescencelifetime was extended in the lipid formulation in comparison to thechemiluminescent system in solution. When applied to porcine skin, ourformulation remained detectable relative to negative and positivecontrols. Initial MTT toxicity testing using HepG2 cells have indicatedthat this formulation is relatively non-toxic. This formulation could beused to image native peroxides present in tissue that may be indicativeof skin disease.
