We describe estimation of total cortisol in serum by HPLC. Incorporation of a new technique to totally automate sample preparation decreases costs. The benefits of HPLC ensure that interferences from other steroids are avoided. The method can also be used to estimate concentrations of other glucocorticoids in serum.
