Abstract

Thrombosis and inflammation have been commonly identified as linked processes. While neutrophils are one of the first responders to areas of inflammation, they are also the first cells to arrive and collaborate with platelets in thrombosis. Recently, the crosstalk between platelets and neutrophils has been recognized in a growing number of pathophysiological processes known as thrombo-inflammation. Neutrophil and platelet functions are tightly regulated by G-protein coupled receptors (GPCRs), because excessive signaling can cause tissue injury and pathological thrombi, respectively. In this chapter, we describe the GPCRs expressed on neutrophils and platelets and their response to thrombo-inflammatory stimuli, as well as the regulators of this coordinated response. We also highlight pharmacological agents and structural studies that may inform future efforts to target these GPCRs therapeutically.
