Abstract

Abstract: A comparative analysis of three independent cell populations isolated from SC7 human embryonic stem cells was carried out. Cell properties were analyzed at the sixth and later passages. The results on growth characteristics and replicative senescence showed that two populations (SC7–MSC-1 and SC7–MSC-2) revealed a low proliferation index, early replicative senescence, and rapid cell death. These two variants could be characterized only in the sixth passage. In contrast, the third cell population, SC7–MSC-3, proliferated actively (proliferation index was high) and got old later, which made it possible to analyze cellular properties at late passages. Thus, the difference in the differentiation potential in SC7–MSC-3 cells between the 6th and 13th passages was found. Karyotypic analysis of these three cell populations revealed differences in karyotypic stability. These populations at the sixth passage differed in the proportion of cells carrying CD90 and CD105 markers, early ESC differentiation markers, and in the differentiation potential in the adipogenic, osteogenic, and chondrogenic directions. These interpopulation differences may be associated with the process of mesenchymal stem cell (MSC) differentiation from embryonic stem cells. Thus, only one population from three independent MSC populations isolated from the SC7 lineage, named SC7–MSC and having status characteristics, corresponded to the definition of a true MSC lineage.
