Abstract
BOD biosensor applications actually increased along with the development of human needs and the progress of science and technology. But in general still predominantly for applications in the field of medical and environmental.A BOD biosensor based on khamir metabolism was developed in order to shorten the measurement time of BOD value for environmental monitoring. The developed biosensor was fabricated by immobilizing khamir on a thin film in agarose matrix with Nafion® acting as the membrane for ion exchange process. The khamir was harvested from the fermentation Rhodotorula mucilaginosa. The film was then attached to gold as the working electrodes. The BOD biosensor was calibrated by using a solution containing the equivalent amount of glucose a standard sample solution. Optimum condition was observed in a waiting measurement time of 20 min at an applied potential of 500 mV (vs. Ag/AgCl). For analysis of glucose, the response time was observed 20 minutes with detection limit 1 mg/l. Good linear range with oxygen standard solution was observed, R2 0.99 and the estimation of measurement error of 2.45%. The observed BOD value by biosensor showed a good comparison with the conventional. Keywords:
