Abstract

Bulb tissue was cultured on MS medium with 0.5 mg 2,4-D and 0.3 mg zeatin/litre under dark conditions. Subculturing was repeated 3 times and calluses transferred either to agitated liquid MS media (1 mg 2,4-D/litre) or to several solid MS media with differing amounts of NAA and benzyladenine. In the liquid media the calluses developed into small, spherical nodules which, after transfer to solid media, developed normally into plantlets. Green plantlets were also obtained with continuous culture on solid media. Culture at 15-20°C was most suitable for growth and regeneration.
