Abstract
Grammatophyllum stapeliiflorum is listed in Appendix II of the CITES. The population of this orchid has decreased due to human activities and germination in nature is low. The tissue culture technique is one solution for propagating orchids using seeds as a source of explants. Pepton is an organic compound that can increase germination and protocorm development because containing amino acid, protein biotin, pyridoxine, thiamin and nitrogen. The research was conducted to determine the effect of peptone on seed germination and protocorm development of G. stapeliiflorum through tissue culture technique. The method used a Completely Randomized Design (CRD) with 6 treatments and 4 replications. The mature seed from sterilized capsule was sown on MS mediumwithout peptone as a control treatment and added with various concentrations of peptone (0.5; 1; 1.5; 2; and 2.5 g/L). The statistical analysis results showed that peptone had a significant effect in accelerating germination. The highest percentage of seed germination (88.59%), protocorm formation (61.02%), and protocorm with SAM and RAM (22.43%) was observed in MS medium containing 2 g/L peptone.
