Abstract
A direct reaction-rate method is proposed for the sensitive determination of uric acid, based on the uricase (Urate Oxidase, EC 1.7.3.3) catalyzed oxidation of uric acid: [formula omitted] Using a platinum electrode which is covered with an immobilized uricase as a thin reaction layer, the disappearance of dissolved oxygen during the enzymatic degradation of uric acid is directly measured at —0.60 V vs. SCE. The initial rate of disappearance of dissolved oxygen is proportional to the uric acid present. By this method, the assay of uric acid in human serum and urine can be performed selectively and rapidly. © 1974, American Chemical Society. All rights reserved.
